Al inicio de las hostilidades se despachó una fuerza de jabos para atacar el aeródromo griego de Alexandrovo. Los Bf 109 equipados con bombas ventrales de 250 Kg lograron destruir 7 aparatos enemigos en el suelo, pero en el primer día el III./JG 27 perdió cuatro de sus aparatos como ya hemos visto. Ese mismo día el Kommodore del JG 27 derribó un Hurricane del Squadron nº 80 de la RAF cerca de Tanagra, al suroeste de Grecia. El 10 de abril los Bf 109 del 6./JG 27 derribaron a 6 bombarderos Blenheim británicos en tan solo 4 minutos de combate sobre Bitjol, como ya hemos visto anteriormente y para el día 14 los enfrentamientos sobre Yugoslavia había cesado trasladándose la acción a Grecia.

El mismo día 14 el Hauptmann Gerlach, de la 6./JG 27, fue derribado y capturado por los británicos, también cayó el Leutnant Hans-Jakob Arnoldy del 4./JG 77, derribó un Hurricane sobre el aeródromo de Larissa alcanzando su victoria número 7 pero fue herido por dos disparos certeros que le alcanzaron el pecho. Su verdugo fue el as canadiense Flt Lt John Mackie’s (7 victorias) del Squadron nº 33 de la RAF. Para el 15 del mismo mes los cazas alemanes ya combaten denodadamente contra viejos conocidos, los pilotos de la RAF.

A lo largo del día 15 se suceden pequeñas batallas aéreas, durante los combates los Bf 109 derriban 7 cazas enemigos, entre griegos y británicos, sin sufrir ninguna pérdida propia al oeste de la ciudad de Trikkala. El Oberleutnant Rödel logra derribar 4 de los aparatos (un Gladiator, un Bloch MB.151 y dos PZL 24), el Oberleutnant Wiesinger y el Leutnant. Börngen se anotan un Gladiator cada uno, además de otro PZL 24 abatido a manos del Oberfeldwebel Schulz. Otro Gladiator fue derribado en combate aéreo por un Bf 109 del 4./JG 27 pilotado por el Leutnant Ernst Börngen el día 15 de abril cerca de Kalambaka. Al día siguiente la caza griega disponía de 21 aparatos operativos, 8 Gladiator, 11 PZL 24 y dos MB.151 sin embargo 18 de ellos resultaron destruidos en un ataque sorpresa de los Bf 109 el día 19. Aunque los griegos evacuaron sus tres máquinas restantes a Eleusis corrieron la misma suerte ya que los Bf 109 del II./JG 27 atacaron esta posición el día 20.

El avance alemán permitió al JG 27 avanzar sus posiciones hasta Ptolemais el día 16. El 18 de abril proporcionó cobertura a una formación de Ju 87 pero en esta misión se perdieron 2 de los cazas al colisionar contra un Ju 87 y un Do 17. Al día siguiente se anotaron el derribo de dos Hurricane más (uno abatido por el Oberleutnant Wiesinger y otro a cargo del Unteroffizier Lippert) sin embargo el aparato de Wiesinger resultó dañado en el combate y tuvo que efectuar un aterrizaje forzoso tras las líneas enemigas cuando realizaron un ataque al suelo en Tanagra. Sobre Tebas el Oberfeldwebel Rockel fue derribado, logró hacer un aterrizaje forzoso pero recibió un fuerte golpe en la cabeza que le provocó una conmoción, los soldados australianos le llevaron a un hospital de Tebas donde fue liberado poco después por tropas alemanas. […]. La suerte del III./JG 27 fue bien distinta, la 8./JG 27 fue enviada para efectuar un ataque sobre las posiciones griegas en el paso de Rupel cuando fueron interceptados los Hurricane del Squadron nº 33 de la RAF. Cuatro de los ocho Bf 109 fueron derribados, entre ellos el Oberleutnant Arno Becker, el tercer Staffelkapitän seguido que había perdido la 8./JG 27. Sin embargo otros pilotos alemanes continuaban aumentando sus cuentas como el caso del Leutnant Fritz Geisshardt de la Stab I./LG 2 que derribó un Hurricane el día 16 y otro el día 26 sobre la isla de Creta con lo que alcanzó los 19 derribos.

El 26 de abril la Stab, II. y III. del JG 27 fueron retirados a Alemania y Polonia para descansar antes de la invasión de Rusia. A principios de mayo se retraso la salida del III./JG 27 que pasó a Sicilia junto con la 7./JG 26 para la ofensiva aérea del Eje contra Malta. Esta unidad voló diversas misiones (escolta de bombarderos, Freire Jagd, Jabo,...) sobre Grecia abatiendo a 5 Hurricane, uno de ellos del Squadron nº 185 de la RAF reclamado por el Gruppenkommandeur Hauptmann Max Dobislav el 15 de mayo. Durante las operaciones sobre Malta se anotaron 5 victorias aéreas entre el 6 y el 20 de mayo, la primera de ellas registrada el día 6 por el Oberleutnant Graf von Kageneck quién derribó al Hurricane Z3057 (pilotado por el Plt. Off. Derdge). La última victoria registrada en ese periodo también fue destruida por von Kageneck, un Hurricane N2673 a los mandos del Plt. Off. Reevesa.
